Key Properties
- A functional phospholipid consisting of a DSPE lipid domain and a free thiol group (-SH) at the terminal end of the head group.
- Amphiphilic structure: hydrophobic saturated stearoyl tails provide stable insertion into lipid bilayers, while the thiol group is exposed on the surface for conjugation.
- Undergoes highly specific reactions with maleimide-functionalized molecules via Michael addition, forming stable thioether bonds.
- Can form disulfide bonds (-S-S-) through oxidation, enabling reversible crosslinking or conjugation.
- Biocompatible, low cytotoxicity, and highly stable in liposomal and lipid nanoparticle formulations.
- Practically insoluble in water; soluble in chloroform, methanol, DMSO, and ethanol.
- Usually appears as a white to pale yellow waxy solid.
- Susceptible to oxidation; recommended storage at −20 °C under inert gas and desiccated conditions.
