Liquid Chromatography – Tandem Mass Spectrometry (LCMSMS)

Liquid Chromatography with tandem mass spectrometry (LCMSMS) combines the separating power of liquid chromatography with the highly sensitive and selective mass analysis of triple quadrupole mass spectrometry.

A sample solution containing analytes of interest first undergoes liquid chromatography separation. The different chemical interaction between the components of the sample, the stationary phase and the mobile phase leads to different migration rates through the LC column. Therefore, a good separation of the sample mixture can be achieved by modifying the stationary phase and mobile phase.

After the separation, the eluent is directed to the triple quadrupole mass spectrometer, where the effluent is nebulized, desolvated and ionized to create charged particles. These charged particles then migrate under high vacuum through a series of mass analyzers (quadrupole) by applying electromagnetic fields. A specific mass/charge precursor ion (or parent ion) can be targeted to pass through the first quadrupole, excluding all other mass/charge ratio particles. The selected mass/charge ions are then fragmented into product ions (or daughter ions) by collision with an inert gas in the collision cell, where the third quadrupole is used to target specific product ion fragments. The resulting isolated product ions are then quantified with an electron multiplier. This transition of ions from the precursor to product ion (also referred to as MS2) is highly specific to the structure of the compound of interest and therefore offers a high degree of selectivity.

LCMSMS combines the separation power of LC and the high selectivity/sensitivity of the triple quadrupole mass spectrometry and offers researchers a robust tool to quantify compounds based on their unique mass/charge (m/z) transitions with high confidence.
