TAMRA CPG 500, 5-Isomer
TAMRA CPG 500 is a controlled pore glass (CPG) solid support used for synthesizing 3’-labeled oligonucleotides. It features the fluorescent dye TAMRA (carboxytetramethylrhodamine), emitting orange-red light (~563-580 nm). It is commonly used in real-time PCR, probe-based assays, and fluorescence experiments requiring dual-labeled oligonucleotides.
Key Features:
- Type: CPG solid support with TAMRA dye
- Pore Size: 500 Å (optimal for oligonucleotides up to 50-mer)
- Fluorescence: Orange-red emission (~563-580 nm)
- Stability: Sensitive to ammonium-containing solutions
- Compatibility: Works with standard DNA/RNA synthesizers
Applications:
- Oligonucleotide Synthesis: 3’-labeling of primers and probes
- Probe-Based Assays: Real-time PCR (TaqMan probes) and FRET experiments
- Diagnostics & Research: DNA sequencing, mutation detection, and genetic analysis
Usage Guidelines:
- Coupling: Standard time depending on the first monomer used
- Deprotection: Use the TAMRA cocktail (tert-butylamine:methanol:water, 1:1:3) for 6 hours at 60°C, then cool to room temperature
Important: Avoid ammonium-based deprotection solutions to prevent irreversible degradation of the TAMRA dye.
