DSPE-Biotin
Full Name: 1,2-Distearoyl-sn-glycero-3-phosphoethanolamine-N-biotin
CAS No.:133695-76-8
Key Properties
- A bioconjugatable phospholipid composed of a DSPE lipid moiety and a biotin group linked covalently on the polar head.
- Amphiphilic: highly hydrophobic distearoyl tails insert stably into lipid bilayers, while biotin is exposed on the surface for molecular recognition.
- Exhibits extremely strong and specific binding affinity to streptavidin/avidin, one of the strongest non-covalent biological interactions.
- High biocompatibility, low toxicity, and good stability in liposomal and nanoparticle formulations.
- Practically insoluble in pure water but soluble in organic solvents including chloroform, methanol, and DMSO.
- Usually appears as a white to off-white solid or waxy powder.
- Recommended storage: −20 °C, desiccated and protected from light.
Applications
- Surface functionalization of liposomes, lipid nanoparticles (LNPs), micelles, and exosomes for biotin–streptavidin capture and detection.
- Immobilization of lipid-based drug carriers onto streptavidin-coated surfaces for biosensing and purification.
- Targeted delivery systems: binding to biotinylated antibodies, peptides, or targeting ligands via streptavidin bridges.
- Cell and membrane labeling for fluorescence imaging, flow cytometry, and immunohistochemistry.
- Purification and isolation of lipid assemblies using streptavidin resins or magnetic beads.
- Used in mRNA, DNA, and siRNA delivery systems to improve targeting and detection of lipid carriers.
