DSPE-Alkyne
Full Name: 1,2-Distearoyl-sn-glycero-3-phosphoethanolamine-N-alkyne
CAS No.: 2933227-70-2
Key Properties
- A functionalized phospholipid consisting of a DSPE lipid backbone and a terminal alkyne (-C≡CH) group on the hydrophilic head.
- Amphiphilic structure with highly hydrophobic saturated stearoyl tails, providing excellent stability in lipid bilayers.
- Reactive alkyne group enables bioorthogonal click chemistry, specifically copper-catalyzed azide-alkyne cycloaddition (CuAAC) with azide-modified molecules.
- Can also be used in strain-promoted cycloaddition with DBCO-containing compounds when copper-free conditions are required.
- High biocompatibility and low toxicity, suitable for biological systems.
- Insoluble in water but soluble in organic solvents such as chloroform, methanol, and DMSO.
- Typically appears as a white to off-white waxy solid or powder.
- Stable under inert atmosphere; recommended storage at −20 °C, protected from light and moisture.
Applications
- Surface modification of liposomes, lipid nanoparticles (LNPs), micelles and exosomes via click chemistry.
- Conjugation with azide-functionalized targeting ligands, peptides, antibodies, and aptamers for targeted drug delivery.
- Labeling of lipid assemblies with fluorescent or radioactive azide probes for in vitro and in vivo imaging.
- Construction of multifunctional theranostic nanoparticles combining therapy, targeting, and imaging.
- Preparation of functional PEGylated lipids and biomimetic membrane systems.
- Used in nucleic acid delivery systems (DNA, mRNA, siRNA) for surface functionalization of delivery vehicles.
