Azide-PEG3-tetrazine is a dual-functional connector used in bioconjugation, featuring azide and tetrazine groups on either side of a PEG3 (triethylene glycol) spacer. The PEG component enhances water solubility. The azide part readily engages with terminal alkynes, BCN, and DBCO through both copper-assisted and copper-free click reactions. The tetrazine end reacts swiftly with terminal alkenes and trans-cyclooctenes through inverse electron demand Diels-Alder (IEDDA) ligation and can also interact with specific strained cycloalkynes.
