TCO-PEG1-Tosyl (axial) is a versatile reagent commonly used in bioconjugation reactions for the covalent attachment of TCO (trans-cyclooctene) to biomolecules such as proteins, peptides, and nucleic acids. The reagent contains a TCO group, which is an alkene with a strained cyclooctene ring structure that undergoes a rapid and selective reaction with a complementary functional group, tetrazine (Tz), in a bioorthogonal “click” reaction.
The TCO-PEG1-Tosyl (axial) reagent also contains a polyethylene glycol (PEG) linker, which can be used to increase the solubility and biocompatibility of the modified biomolecules. The PEG linker can also act as a spacer, preventing steric hindrance between the TCO group and the biomolecule of interest, and improving the accessibility of the TCO group for the tetrazine functional group.
Additionally, the TCO-PEG1-Tosyl (axial) reagent contains a tosyl (p-toluenesulfonyl) leaving group, which allows for the efficient conjugation of the reagent to the biomolecule of interest via nucleophilic substitution reaction with primary amines. This reaction is commonly used to modify lysine residues in proteins, resulting in the formation of a stable amide bond.
