LumiTracker ER Red (also known as ER-Tracker™ Red) is a red-fluorescent dye made from BDP TR dye coupled with Glibenclamide (Glyburide). Glibenclamide binds to sulphonylurea (SUR) receptors on ATP-sensitive potassium channels (KATP), commonly found on the endoplasmic reticulum (ER). This selective binding makes it ideal for live-cell ER imaging. Although some staining remains after formaldehyde fixation, LumiTracker ER Red is not suitable for use on pre-fixed cells.
Key Features:
- ER-Specific Binding: Targets KATP channels through Glibenclamide’s selective binding.
- Live-Cell Compatibility: Allows real-time ER imaging without harming cell health.
- Fixation Limitation: Staining partially survives formaldehyde fixation but is unsuitable for pre-fixed cells.
LumiTracker ER Red Applications:
- ER Visualization: Provides clear ER staining for live-cell microscopy.
- Cell Biology Studies: Helps explore protein synthesis, folding, and ER-related processes.
How to Use:
- Preparation: Dissolve the dye in DMSO to prepare a stock solution. Dilute to working concentrations based on your experiment.
- Storage: Keep at -20°C, protected from light. Avoid repeated freeze-thaw cycles.
Note: Since sulphonylurea receptor expression can vary between cell types, verify staining performance for your specific experiment.
