DSPE-Polysarcosine-Azide20
Full Name: 1,2-Distearoyl-sn-glycero-3-phosphoethanolamine-N-poly(sarcosine)-azide (average 20 sarcosine repeating units)
Abbreviation: DSPE-pSar20-N₃
Key Properties
- A trifunctional amphiphilic lipid conjugate consisting of a hydrophobic DSPE bilayer anchor, a hydrophilic polysarcosine (pSar20) stealth spacer, and a reactive azide (-N₃) end group.
- The polysarcosine segment provides excellent antifouling performance, high hydrophilicity, good biocompatibility, and low immunogenicity, acting as a biodegradable and non-immunogenic alternative to PEG.
- The azide group enables highly efficient bioorthogonal copper-catalyzed click chemistry (CuAAC) with alkyne-modified molecules, or copper-free SPAAC with DBCO-modified ligands under mild physiological conditions.
- Can stably insert into lipid bilayers while exposing reactive azide groups on the nanoparticle surface.
- Soluble in organic solvents including chloroform, methanol, and DMSO; dispersible in aqueous buffers.
- Typically appears as a white to off-white solid or waxy powder.
- Storage: −20 °C, desiccated, protected from light and moisture to maintain azide stability.
Applications
- Used as a reactive stealth lipid for surface functionalization of liposomes, lipid nanoparticles (LNPs), micelles, and exosomes.
- Enables covalent conjugation with alkyne/DBCO-modified targeting ligands, antibodies, peptides, aptamers, and fluorescent or radioactive probes.
- Reduces nonspecific protein adsorption and prolongs blood circulation time of nanocarriers in drug and nucleic acid (mRNA, siRNA, DNA) delivery.
- Applied in targeted drug delivery, vaccine development, and in vitro/in vivo molecular imaging.
- Serves as a PEG alternative to avoid accelerated blood clearance (ABC) and immune responses associated with PEG lipids.
- Used in biomembrane engineering and surface modification of biomaterials for improved biocompatibility and targeting ability.
