TAMRA Phosphoramidite, 6-Isomer
A fluorescent dye with a phosphoramidite group for synthesizing 5’-labeled oligonucleotides. It emits in the orange spectrum (maximum at 563 nm) and is widely used in molecular biology assays, particularly in qPCR and fluorescence-based diagnostics.
Key Features:
- Dye Type: TAMRA (Carboxytetramethylrhodamine) from the rhodamine family
- Reactive Group: Phosphoramidite for 5’-labeling of oligonucleotides
- Emission: Orange fluorescence (~563 nm)
- Applications: DNA/RNA synthesis, molecular diagnostics, qPCR
- Compatibility: Works with standard DNA synthesizers
Applications:
- 5’-Labeling of Oligonucleotides: For quantitative PCR and fragment analysis (e.g., microsatellite markers).
- FRET-Based Probes: Acts as a FRET acceptor in TaqMan probes when paired with fluorescein (FAM).
- Dual-Labeled Probes: Used with Fluorescein dT Phosphoramidite or FAM CPG for TaqMan assays.
Recommended Usage:
- Coupling Time: 7.5 minutes.
- Deprotection Method: Use TAMRA cocktail (tert-butylamine:methanol:water, 1:1:3) for 6 hours at 60°C, then cool to room temperature.
Important: Avoid ammonium-containing deprotection solutions (aqueous ammonium or AMA), as they cause irreversible degradation of the TAMRA dye.
