ProteOrange Protein Gel Stain (5000X)
ProteOrange is a highly sensitive fluorescent dye for staining proteins in polyacrylamide gels, offering about 10 times higher sensitivity than Coomassie Blue. It can detect as little as 3 ng of protein per band.
Key Features:
- Sensitivity: Detects protein bands as low as 3 ng.
- Concentration: Supplied as a 5000X solution in DMSO, enough for approximately 200 mini-gels per 1 mL.
- Compatibility: Works with SDS-PAGE gels, binding selectively to proteins even in complex samples.
- Ease of Use: Requires simpler staining procedures compared to silver staining, with no need for fixation or extensive washing.
Application Protocol:
- Preparation:
- Ensure the dye stock is free from precipitates. If needed, warm at 70°C and mix well.
- Working Solution:
- Dilute the 5000X stock in 7.5% acetic acid (e.g., 5 µL ProteOrange in 25 mL acetic acid).
- Staining:
- After electrophoresis, immerse the gel in the working solution. Incubate in the dark for 20–60 minutes.
- Rinsing:
- Rinse the gel briefly with 7.5% acetic acid for 30 seconds.
- Visualization:
- View the stained gel using a UV transilluminator (312–365 nm).
ProteOrange is ideal for protein visualization in biochemical research and is available from various suppliers.
