4-Formyl-2-nitrophenyl-β-D-glucopyranosiduronic acid methyl ester 2,3,4-triacetate CAS:148579-93-5 is a β-glucuronic acid-based enzymatic cleavable linker. It is developed in an extension of the peptide-based linker strategies to provide high ADC stability. Facile release of the active drug can be achieved through cleavage of the b-glucuronide glycosidic bond by the lysosomal enzyme b-glucuronidase (GUSB).
β-glucuronidases, found exclusively in the lysosomal compartment of the cell, are a class of hydrolytic lysosomal enzymes that degrade β-glucuronic acid residues into polysaccharides.
