The tetrazine group readily reacts with TCO-tagged compounds to form a stable covalent bond through a ultrafast bioorthogonal reaction – TCO-tetrazine ligation. The hydrophilic PEG spacer can increase aqueous solubility of the biotinylated molecules and help minimize steric hindrance and provide extra flexibility with the binding to stravidin and avidin molecules.
