NI-57 CAS:1883548-89-7 is a potent BRPF bromodomain inhibitor that binds BRPF1B with a KD of 31 nM (ITC) and BRPF2 with a KD of 108 nM (ITC) and BRPF3 with a KD of 408 nM (ITC) ). NI-57 is highly selective for other non-class IV bromodomains, including BET, as measured by biophysical and biochemical methods. The closest off-target effect was against BRD9 (32-fold selectivity). Inhibits RANKL-induced differentiation of primary murine bone marrow cells and primary human monocytes into osteoclasts.
