Methyltetrazine-Sulfo-NHS ester
This product is a water-soluble, heterobifunctional reagent that combines a methyltetrazine group with a sulfo-NHS ester. The methyltetrazine enables ultrafast, copper-free ligation with trans-cyclooctene (TCO) via inverse electron-demand Diels–Alder (IEDDA) chemistry, while the sulfo-NHS ester reacts efficiently with primary amines to form stable amide bonds. Its sulfonated structure provides excellent aqueous solubility, making it especially ideal for direct bioconjugation of proteins, antibodies, and other biomolecules.
Key Features and Benefits
-
Methyltetrazine Group: Supports rapid, bioorthogonal ligation with TCO under physiological conditions.
-
Sulfo-NHS Ester: Couples selectively with amines (lysines, N-termini) to form amide linkages.
-
Water-Soluble: Sulfonated ester improves reactivity in aqueous systems.
-
Dual Reactivity: Enables orthogonal labeling strategies combining IEDDA and amide coupling.
-
High Purity: Produced to ≥95% purity for consistent performance.
Applications
-
Bioconjugation: Links tetrazine and TCO-modified biomolecules.
-
Protein and Antibody Labeling: Attaches tetrazine tags to proteins directly in aqueous buffers.
-
Probe Development: Builds dual-functional imaging and diagnostic probes.
-
Drug Delivery: Serves as a linker for targeted therapeutic systems.
Storage and Handling
-
Storage: Store at –20 °C in a sealed, moisture- and light-protected container.
-
Handling: Dissolve freshly in aqueous buffer; avoid repeated freeze–thaw cycles.
This product integrates ultrafast tetrazine reactivity with amine coupling and water solubility, specifically making it a powerful reagent for protein labeling, probe design, and advanced bioconjugation.
