IR 750 DBCO (IRDye® 750 DBCO analog) is a water-soluble, bright, near-IR fluorescent probe that is pH-insensitive from pH 4 to pH 10. Its excitation band is ideally suited for the 633 nm laser lines or dye-pumped laser. IR 750 dye is spectrally similar to other 750 dyes used for flow cytometry, microscopy and other applications. A significant advantage to using long wavelength dyes such as Cy7® Dye or Alexa Fluor® 750 dye over other fluorophores is the low autofluorescence of biological specimens in this region of the spectrum.
IR 750 DBCO is mainly used for imaging of azide-containing biomolecules without the need for copper catalyst. It reacts with azides via a copper-free “click chemistry” reaction to form a stable triazole and does not require Cu-catalyst or elevated temperatures. In applications like staining the surface of live cells or if there are concerns about native protein function loss in the presence of copper IR 750 DBCO is an excellent alternative to copper requiring fluorescent alkynes.
IR 750 DBCO reagent is not suitable for staining intracellular components of fixed and permeabilized cells due to high backgrounds.
