DODMA (1,2-dioleyloxy-3-dimethylaminopropane) CAS:104162-47-2, an ionizable cationic lipid with a protonable tertiary amine head group, is considered a non-viral vector for gene therapy. DODMA is very effective at encapsulating nucleic acids because of its ability to temporarily lower pH. Shows a positive charge at low pH. DODMA efficiently encapsulates nucleic acids to form lipid complexes during synthesis by lowering the physiological pH, which readily passes through cell membranes. These properties make DODMA ideal for transfection and drug delivery applications. It has been used in the composition of liposomes to be made into stable nucleic acid lipid particles that can encapsulate plasmid DNA, siRNA/miRNA and small molecules of CRISPR/Cas9.
