IR 750 Alkyne (IRDye® 750 Alkyne analog) is a water-soluble, bright, near-IR fluorescent probe that is pH-insensitive from pH 4 to pH 10. Its excitation band is ideally suited for the 633 nm laser lines or dye-pumped laser. IR 750 dye is spectrally similar to other 750 dyes used for flow cytometry, microscopy and other applications. A significant advantage to using long wavelength dyes such as Cy7® Dye or Alexa Fluor® 750 dye over other fluorophores is the low autofluorescence of biological specimens in this region of the spectrum.
The near-IR fluorescent IR 750 Alkyne can be reacted with azides via a copper-catalyzed click reaction (CuAAC) to form a stable triazole bond. Click chemistry-based detection of azide-tagged biomolecules in complex biological samples can be done with unprecedented sensitivity due to extremely low background.
A probe for copper-less detection azide-tagged biomolecules, IR 750 DBCO, is also available for application where the presence of copper is not acceptable.
