HMRhoNox-M (LysoRhoNox) is a Fe(II)-selective fluorescent probe based on the N-oxide-controlled spirocyclization of tetramethyl-hydroxymethyl rhodamine. Without Fe(II), it remains in a non-fluorescent, spirocyclic form, showing minimal fluorescence in aqueous buffers and at physiological pH. Adding Fe(II) induces a 60-fold fluorescence increase at 575 nm as the probe converts to an open, fluorescent form.
Key Features:
- High Selectivity: Targets Fe(II) specifically, avoiding interference from Fe(III), alkali, and alkaline earth metals.
- Dose-Dependent: Fluorescence intensifies in line with Fe(II) levels.
- Lysosomal Targeting: Permeates cells and localizes in lysosomes, ideal for tracking iron fluctuations in live cells.
Applications:
- Iron Detection: Delivers a strong, specific fluorescence response for Fe(II).
- Live Cell Imaging: Tracks real-time iron uptake, including transferrin-induced Fe uptake.
- Biological Research: Aids in studying iron’s role in cellular health and disease.
HMRhoNox-M is a powerful tool for exploring cellular iron dynamics and understanding iron-related processes in health and disease.
