Overview:
DiO, also called DiOC18(3), is a green fluorescent lipophilic carbocyanine dye. It has an excitation max at 487 nm and an emission max at 501 nm. DiO integrates easily into cell membranes, making it perfect for anterograde and retrograde neuronal tracing in live and fixed tissues. It uniformly labels neurons by diffusing through the plasma membrane.
In intact tissues, DiO stays within labeled cells, though some transfer might occur if the membrane gets disrupted, such as during tissue sectioning. Often paired with DiI in dual-color studies, DiO diffuses more slowly across membranes compared to DiI.
