Di-8-ANEPPS, CAS Number: 157134-53-7
Overview:
Di-8-ANEPPS is a fast-responding, voltage-sensitive dye from the ANEP family. It binds to cell membranes and fluoresces only when electrical potential changes are detected. Therefore, with a 2-10% fluorescence change per 100 mV, it efficiently tracks transient voltage shifts in neurons, cardiac cells, and intact brain tissue. Moreover, the dye shifts its excitation spectrum in response to voltage changes, thereby enabling ratiometric measurements, which provide more accurate voltage tracking.
Thanks to its higher lipophilicity, Di-8-ANEPPS stays in the outer membrane longer, making it perfect for long-term studies. It also works well as a plasma membrane marker.
Excitation/Emission Maxima:
- 498/713 nm in methanol
- Blue-shifted in cell membranes
Applications:
- Tracks real-time membrane potential in excitable cells.
- Used in neuroscience and cardiac research.
- Enables ratiometric voltage measurements.
- Serves as a plasma membrane label for extended experiments.
Usage:
Add directly to cultures with Pluronic® F-127 or through retrograde labeling. Start at 5-10 μM and adjust as needed.
Storage:
Keep at -20°C, protected from light and moisture.
