Sulfo-Cyanine5 hydrazide, a sulfated variant of Cyanine5 dye, is highly soluble in water, thanks to its structure which contains two negatively charged sulfo groups. Its spectral characteristics make it a far-red fluorescent dye analogous to Cy™5.
Hydrazides are effective in reacting with aldehydes and ketones to produce hydrazones, making this compound suitable for combining with carbonyl biomolecular derivatives.
The reaction occurs under water-based conditions, a critical factor when dealing with antibodies and proteins. The Cys-diol groups present in glycosylated proteins and antibodies can be oxidized to form dialdehydes, while cysteine in proteins can be enzymatically transformed into formyl glycerol, serving as reactive groups for conjugation with sulfo-Cyanine5 hydrazide. The carboxyl groups of aspartic and glutamic acids in proteins and peptides can also be linked with sulfo-Cyanine5 hydrazide with the help of activating agents like carbodiimide (EDAC) or methyl morpholine (DMTMM) derivatives.
