Aminolink phosphoramidites are used for oligonucleotide synthesis. DMS(O)MT is an improved alternative to traditional MMT with special protecting groups. DMS(O)MT amino linkages are fully compatible with standard coupling, deblocking, and purification protocols. However, the DMS(O)MT amino linker has higher stability and better oligonucleotide recovery, so the DMS(O)MT group is more suitable for column purification of DMT-on and amino oligonucleotides. In the case of column purification, DMS(O)MT protection increases oligonucleotide yields by 2-3 fold, making this reagent a unique solution for high-throughput production of amino oligonucleotides.
