Cy5 Azide NHS Ester is a bright, far-red fluorescent dual-labeled click chemistry probe with absorption and emission spectra nearly identical to Alexa Fluor® 647, CF® 647 dyes, or any other Cyanine5-based fluorochromes for simultaneous ligation of azide group and far-red fluorescent Cy5 labeling. Conjugates prepared with Cy5 azide NHS esters can react with terminal alkynes via a copper-catalyzed click reaction (CuAAC). It also reacts with strained cyclooctynes to form stable triazoles via a copper-free “click chemistry” reaction and does not require copper catalysts or high temperatures. Cy5 dye is a water-soluble, pH-insensitive, pH 4 to pH 10, far-red fluorescent probe with excitation well suited for 633 nm or 647 nm laser lines.
