BI-9564 CAS:1883429-22-8 is a potent inhibitor of BRD9. BI-9564 binds BRD9 with a higher affinity (KD of 14 nM, ITC) than BRD7 (KD of 239 nM, ITC), is completely negative for BET family members, and shows cellular activity via FRAP 0.1 µM and 1 µM on BRD9 and BRD7, respectively. BI 9564 is a selective inhibitor of the bromodomains of BRD9 and BRD7 (Kds = 14.1 and 239 nM; IC50s = 75 nM and 3.4 µM), showed cellular viability by semi-quantitative FRAP assay, ~90% inhibition of BRD9 and BRD7 by 0.1, respectively μM and 1 μM. Inhibits proliferation of AML cell lines in vitro and reduces AML tumor xenograft growth in vivo.
