BI-2536 is a 2-aminopyrimidine-containing Plk1 inhibitor (IC50 = 0.83 nM). BI 2536 has IC50s of 0.83 and 25 nM for PLK1 and BRD4 inhibitors, respectively. BI 2536 also blocked the activity of Plk2 and Plk3 to a lesser extent with IC50s of 3.5 nM and 9.0 nM, respectively. BI-2536 inhibits IFNB (encoding IFN-α) gene transcription. BI-2536 induces mitotic arrest and apoptosis in multiple human cancer cell lines and drives regression of human tumor xenografts in nude mice.
