Azido-PEG8-Val-Cit-PAB-PNP from Axispharm is a linker for antibody-drug conjugation (ADC). The Val-Cit linker can be cleaved by cathepsin B. It is also a PEG-based PROTAC linker that can be used for PROTAC synthesis. PROTAC contains two distinct ligands linked by a linker; one is a ligand for the E3 ubiquitin ligase and the other is a ligand for the target protein. PROTACs utilize the intracellular ubiquitin-proteasome system to selectively degrade target proteins, and since this enzyme is only present in the lysosome, the ADC payload will be released only in the cell. The azide group will react with DBCO, BCN, or other alkynyl groups via click chemistry, and the PEG spacer increases water solubility.
