Overview:
Di-4-ANEPPS is a fast-responding, voltage-sensitive dye from the ANEP family. It binds to cell membranes and fluoresces in response to electrical potential changes, making it ideal for tracking membrane potential in neurons, cardiac cells, and other excitable cells. Its quick optical response detects transient voltage shifts, causing a fluorescence change of 2-10% per 100 mV. It also enables ratiometric measurements by shifting its excitation spectrum with membrane potential changes.
Because Di-4-ANEPPS is rapidly internalized by cells, it is most effective for short-term studies. For longer retention, you can opt for Di-8-ANEPPS, which has higher hydrophobicity. In addition to its role in membrane potential tracking, Di-4-ANEPPS also acts as a plasma membrane and endocytosis marker.
