APDye 430 hydrazide (Alexa Fluor 430 Equivalent) is a fluorescent dye featuring an excitation maximum at 430 nm and an emission maximum at 542 nm. Among the limited number of dyes that absorb between 400 nm and 450 nm, APDye 430 stands out. The fluorescence of APDye 430 is photostable and does not exhibit pH sensitivity over a wide pH range.
This product is derived from APDye 430 through a hydrazide reaction. Hydrazides are known for their efficient reaction with aldehydes and ketones, leading to the formation of hydrazones. As such, this compound is ideal for conjugation with carbonyl derivatives of biomolecules.
The reaction can take place in aqueous conditions, a significant advantage when working with antibodies and proteins. The cys-diol groups present in the sugars of glycosylated proteins and antibodies can be oxidized to dialdehydes, and cysteine in proteins can be enzymatically converted to formyl glycerol (i.e., reactive groups for conjugation with hydrazides). The carboxyl groups of aspartic and glutamic acids in proteins and peptides can also undergo conjugation with hydrazides, provided that activating agents such as carbodiimide (EDAC) or methyl morpholine (DMTMM) derivatives are present.
