Sulfo-SMCC CAS:92921-24-9 is a non-cleavable and membrane-impermeable crosslinker containing one amine-reactive N-hydroxysuccinimide (NHS ester) and one sulfhydryl-reactive maleimide group. NHS esters react with primary amines at pH 7-9 to form stable amide bonds. Maleimide reacts with sulfhydryl groups at pH 6.5-7.5 to form stable thioether linkages. The maleimide groups of Sulfo-SMCC and SMCC are exceptionally stable at pH 7.5 due to the cyclohexane bridge in the spacer arm. Because it contains a hydrophilic sulfonyl moiety, Sulfo-SMCC has a solubility of up to ∼10 mM in water and many common buffers, thereby avoiding the use of organic solvents that can disrupt protein structure.
