Azido-PEG8-t-Boc-hydrazide linker contains an azide group and a Boc-protected hydrazide. The hydrophilic PEG spacer can increase solubility of a compound in aqueous media. The azide group readily reacts with alkyne, BCN, DBCO through Click Chemistry to yield a stable triazole linkag. After de-Boc protection, the free hydrazide group can be reacted with an aldehyde or ketone to form a hydrozone bond that are reversible under acidic conditions. The reversibility of spacer has been widely utilized to for payload traffiking and release.
